Creative Bioarray Has Developed Cell Immortalization Services to Support Potential Drug Development

Currently, Creative Bioarray has developed several HT® Cell Immortalization Kits for immortalizing mammalian cells in culture: 

Viral genes, such as EBV, HPV-16 E6/7 genes, and SV40 T antigen, are used to induce immortalization. In most cases, viral genes are immortalized by inactivating tumor suppressor genes (p53, Rb, etc.) that can induce a replicative senescence state in cells.

SV40 T antigen has been shown to be the simplest and most reliable reagent for transforming many different cell types in culture, and the mechanism of SV40 T antigen in cell immortalization has been intensively studied. Recent studies have also shown that the SV40 T antigen can induce telomerase activity in infected cells.

A recently discovered method of cell immortalization is through the expression of TERT, especially for cells that are most affected by telomere length. This protein is inactive in most somatic cells, but when hTERT is exogenously expressed, cells are able to maintain sufficient telomere length to avoid replicative senescence. Analysis of several telomerase-immortalized cell lines has confirmed that cells maintain a stable genotype and retain key phenotypic markers.

Co-expression of the hTERT catalytic subunit with p53 or RB siRNA immortalizes primary human ovarian epithelial cells, providing a more realistic normal cell model with well-defined genetic background.
