As mass spectrometry advances, the identification of proteins and peptides is no longer cumbersome and time consuming. Creative Proteomics provides N-terminal sequencing analysis of therapeutic proteins, monoclonal antibodies and protein vaccines by Edman and mass spectrometry. In protein Edman N-terminal sequencing, the protein was first modified with phenyl isothiocyanate (PITC). Subsequently, the derivatized terminal amino acid is removed by acid cleavage in the form of a phenylhydantoin (PTH) derivative, and the new α-amino group on the next amino acid is now available for the next round of reaction with PITC. Therefore, the protein sequence is being analyzed by the severe reaction of PITC addition, and one cleavage of one PTH at a time for analysis. Suppose the reaction are completely effective, the method can sequence the entire amino acid from the N-terminal end. Although less efficient than mass spectrometers, N-terminal Edman sequencing still has the advantage of protein analysis, while other analytical methods are not readily available.

 

De Novo Peptides/Proteins Sequencing Service is important to study the biological function of the peptide/protein. However, a complete characterization of peptides/proteins, including post-translational modifications (PTMs), sequence mutations and variants, is very challenging. There are two approaches to determine the sequence of peptide/protein by mass spectrometry: database search and de novo sequencing. Database search approach compares acquired mass spectra to a database of known protein sequences to identify the protein sequences. De novo sequencing is a process in which amino acid sequences are directly interpreted from tandem mass spectra without the assistance of a database.

 

Peptide Mass Fingerprinting (PMF) is an analytical technique for identifying the unknown protein. Proteins to be identified is first digested into peptides, whose absolute mass can be accurately analyzed by a mass spectrometer, such as MALDI-TOF or ESI-TOF.
