Absolute quantification, also known as AQUA analysis, is a targeted quantitative proteomics technology with powerful efficacy and is increasingly used in a variety of quantitative proteomics studies. The AQUA strategy is used for absolute quantification (AQUA) of proteins and their modified states. The synthesized peptide acts as an ideal internal standard together with the incorporated stable isotope to mimic the natural peptide formed by proteolysis. These synthetic peptides can also be prepared by chemically identical covalent modifications (e.g., phosphorylation, methylation, acetylation, etc.) to naturally occurring post-translational modification. The absolute level of proteolytic and post-translationally modified proteins is then accurately and quantitatively measured using such AQUA internal standard peptides by using selected reaction monitoring assays in a tandem mass spectrometer. Advances in biological mass spectrometry have led to the development of a number of strategies for quantifying intracellular protein expression levels. In addition to the measurement of protein expression accomplished by differentially incorporating stable isotopes into cellular proteins, absolute quantification is a useful method in proteomic analysis.
